Stand-over is usually meant to be the amount of room between the top tube and the crotch when you're standing over the bike, dismounted. For road bikes, this should generally only be about 1-2 inches. For mountain bikes, it can vary depending on whether you're riding a hard tail or a full suspension but, 3 inches plus is a good rule of thumb. Stand-over is of less consequence on a road bike, where presumably you'll not be dismounting as much, and not in as potentially dangerous circumstances as you would on a mountain bike. It's more important to get a proper fit from handlebar to seat on a road bike.
